Description
The main goal of this proposal is to fabricate a novel multimodal nanoplatform, based in ferrite phases of radiopaque elements dopped with fluorescent cations, for their use of multimodal contrast agents (MCAs), which are designed specifically for their use in the labeling and therapeutic monitoring of genetically modified regulatory T lymphocytes (CAR-T) for the treatment of autoimmune diseases.
For this purpose, these multimodal platforms should have the ability to generate contrast in magnetic resonance imaging, X-ray computed tomography, and fluorescence microscopy." In doing so, we will develop a new strategy to synthesize these multimodal nanoplatforms which consists in the laser ablation of massive precursorsimmersed into a solvent, which contains hydrophilic functionalizing agents.
This research will be coupled to in situ study of the laser fabrication process. We will analyse the structure, composition, colloidal stability, cytotoxicity and ability of these nanoparticles to produce contrast in MRI, fluorescence microscopy and X-ray imaging by using a battery of analytical techniques.
Finally, we will target these MCAs toward specific biological targets of those autoimmune diseases, selected for study.
