Offer Description
The project aims to study the allosteric modulations that occur within the oligomers constituted by mu-opioid receptors and other G protein-coupled receptors. These complexes are responsible of opioid analgesic effects as well as with the phenomena of tolerance and addiction associated with the continued use of opioids. Therefore, the postdoctoral candidate will conduct research in biochemistry, pharmacology and molecular biomedicine of G protein-coupled receptors (GPCRs) oligomers involved in analgesia, tolerance and addiction. The candidate will focus on the allosteric interactions that occur within these complexes which make them valuable tools for drug development. The candidate will carry out experimental laboratory work, as well as data analysis, discussion of results and proposal of new experiments and approaches. He will be asked to obtain and analyze optical and confocal microscopy images and to develop radiochemical and immunochemical assays. The postdoctoral candidate is expected to work with other predoctoral members of the research team and to mentor them.
The candidate will work in the laboratory of Molecular Neuropharmacology at the Faculty of Biology of the University of Barcelona

- In vitro and cell molecular biology techniques: radioligand binding assays, bioluminescent and fluorescent resonance energy transfer (BRET/FRET) and proximity ligation assays, protein purification techniques, enzymatic activity assays, PCR/qPCR, DNA cloning. Cell culture techniques, cell signaling pathway determinations as G protein activation, cAMP accumulation or ERK1/2 phosphorylation and fluorescent and confocal microscopy.
- Animal models: behavioral assays (B, C and D functions (FELASA)).
- Strong knowledge of standard bioinformatic data resources and scientific software such as GraFit, GraphPadPrism, ImageJ
The candidate will need to have at least 3 years of postdoctoral experience and to demonstrate expertise in studying allosteric interactions between receptors. Specifically, the candidate needs to have proficiency and in performing radiochemical assays for affinity determinations and BRET/FRET and proximity ligation assays for detecting protein-protein interactions applied to receptors. Moreover, she/he will need to demonstrate experience in performing assays to determine cell signaling pathway activations using BRET/FRET methods, label-free and HTRF assays, WesternBlot or immunoassays. She/he must have expertise in confocal microscopy, cell culture and in working with animal models.
The candidate must have publications in international journals on GPCRs in general, and in opioid receptors in particular.
Knowledge of Spanish and English is required.
