Offer Description

In the framework of the AIRC cancer research project lines, the research activity will consist in the development of a 3D substrate that mimics the characteristic structures of the brain with variable rigidity and with studies of mechanosensing proteins in hypoxia. Technologies may include hydrogel polymerization, electrospinning and particulate leaching. In particular, our studies are focused on: (i) identifying the response of glioma stem cells as a function of 3D substrates with different rigidity; (ii) identifying the molecular and biological responses to Hippo effectors YAP/TAZ acting as on–off mechanosensing switches by sensing modifications in extracellular matrix (ECM) composition and mechanics, and the molecular mechanism by which spreading and focal adhesion formation through YAP occurs as well as modulating the YAP transcriptional activity and iii) Relationship between 3d structures of different rigidity and YAP in glioma stem cells in hypoxia. The information gathered by this project will be instrumental in modeling the design of new regenerative strategies proposed by the project. These systems will provide models both for studying mechanotaxis mechanisms and for identifying potential new treatments.

Master’s Degree in (Bio-) Engineering, Biotechnology, Biological Science, and Molecular and Cellular Biology or related subjects;

Specific Requirements

• The applicant must have prior working experience documented by scientific publications, with cell culture, in molecular and cellular biology, including functionality assessment through cellular and molecular biological assay (Western Blot, cell viability, etc). • The applicant must show, excellent molecular biology and live cell microscopy skills and/or experience in cell transfection, quantitative RT-PCR, Electrophoresis, Nucleic Acid extraction, Protein extraction. • Experience documented on the capability to work with biomaterials and fabrication of micro-nano structures would be advantageous. • On-board experience will be positively evaluated. • The ability to work independently and as part of a team, excellent communication, organisational, problem solving skills and writing scientific papers are essential. • Experiences with the Hippo effectors YAP/TAZ, its translocation and relative pathways will be considered additional beneficial skills of the prospective candidate • Good knowledge of English (for foreign candidates). • Basic knowledge of Italian (for foreign candidates). • Preferred title: PhD in Biochemical and Biotechnological Sciences or related subjects.
