Project title: "Dynamics of the OXPHOS system in cellular senescence”. Project is carried out within the “Sonatina” programme of the National Science Centre.


Project description:
Mitochondrial OXPHOS system is a set of five large multiprotein complexes that constitute the “heart” of mitochondria. They drive energy production and metabolism. Interestingly, OXPHOS complexes are not necessarily degraded as a whole but individual elements can be exchanged when damaged. We know OXPHOS can change in response to stress. Yet, what happens to the OXPHOS system, particularly how it becomes repaired, remains enigmatic during a cell stress response known as cellular senescence. We aim to better understand how the OXPHOS system is maintained when cell enter senescence, cellular state associated with increased inflammation and resistance to cell death. We use biochemical and proteomic tools to track OXPHOS turnover patterns and identify mitochondrial protein quality control mechanisms
that take care of the OXPHOS machines in senescent cells. We are also interested to establish how OXPHOS quality control impacts on the cell phenotypes that contribute to senescence cells’ pathological role in ageing. Accomplishing the goals of the project will contribute to our knowledge on how cells respond to stress as well as directs us towards novel anti-ageing (anti-senescence) strategies.

Key responsibilities of candidates:
1. Participate in experimental design;
2. Conduct research experiments (independently and in collaboration with the supervisor) using culture of human cells; basic biochemical methods such as Western blotting, analysis of native complex status with Blue Native PAGE Western blotting, preparation of samples for proteomic analysis; genetic engineering with CRISPR/Cas9 system;
3. Participate in data analysis and dissemination of results among the scientific and broader community.
 

Requirements of candidates:
Bachelor's degree in Biology/Biochemistry/Biotechnology or a related field;
