Offer Description
Our team is interested in the intracellular metabolism of pathogenic mycobacteria and in particular the role and degradation of intracytoplasmic lipid inclusions (ILIs) formed during the infection process. The aim of this project, funded from januray 1st, 2025 to December 31st, 2027 by the Agence Nationale de la Recherche, will focus on the identification of proteins involved in the degradation of these ILIs using APEX2 technology.
The recruited person will be able to
-Generate genetic constructs and recombinant strains used in in vitro models of ILI consumption.
-The biotinylated proteins by APEX 2 will be purified and analyzed by mass spectrometry.
-The proteins identified will be produced in recombinant form and biochemically characterized
-Their physiological role will be studied by generation of deletion strains of identified genes. The ability to consume lipids will then be studied in in vitro model using TLC analysis.
This work will be carried out in the Laboratory of Engineering Macromolecular Systems as part of the Lipolysis and Bacterial Pathogenesis team led by Stéphane Canaan.
The team currently comprises 6 permanent staffs, 4 PhD students and 1 post doc. All the skills required to successfully complete the project are mastered by the various members of the team, who will be able to help the recruited person.

-Mastery molecular biology techniques (PCR, cloning, transformation, construction of deletion mutants)
-Expertise in protein biochemistry (production, purification and biochemical characterization of recombinant proteins)

-Know how to culture and infect eukaryotic cells
-Work in complete autonomy
-Ability to formulate the results obtained and write articles in English
M. tuberculosis, the causative agent of tuberculosis (TB), has the ability to store triglycerides (TAG) in the form of Intracytoplasmic Lipid Inclusions (ILI) within foamy macrophages or in caseous center during granuloma formation. Once gorged with lipids, the bacillus may persist in dormancy phase, allowing latent TB to develop. When tuberculosis reactivation occurs, degradation of the stored TAGs facilitates the spread of the bacillus throughout the body. Using genetic, biochemical, proteomic, microbiological and cellular approaches, Delirious will identify the mycobacterial proteomes involved in the ILI degradation and study their impact on virulence using M. abscessus as model. This original project will provide unprecedented information on the life cycle of pathogenic mycobacteria, providing inspiration for innovative therapeutic strategies to combat mycobacterial related diseases.
