We seek a highly motivated and enthusiastic PhD student for a period of four (4) years at the Department of Molecular Biology, UMCS Lublin, to join the research project aimed at deciphering the ribosome inactivating proteins action at molecular and cellular levels, financed by the NCN (National Science Centre).

About the project

PROJECT OVERVIEW

Some of the most toxic proteins known in nature are the Ribosomal Inactivating Proteins (RIPs). The current accepted model of RIP action depicts that RIPs depurinate (remove one adenine base from the rRNA at the sarcin-ricin loop, SRL) the 60S rRNA, leading to protein biosynthesis inhibition regarded as the main trigger of cell death. However, despite many years of research, the mechanism of RIP toxicity at the cellular level is still obscure. Thus, the project is focused on deciphering the mechanism of action of RIPs using structural/biophysical and molecular biology approaches, as well as analyzing various members of the RIP family.

The project is to bring new light in understanding of the molecular and cellular mechanisms of RIP toxicity towards eukaryotic cells, an issue that has never been solved so far. RIPs represent a hope for designing effective cancer-targeted therapy tools, now limited mainly due to severe side-effects, such as vascular leakage syndrome. Our project will provide unique opportunities not only to understand the biology of RIPs, but also, by development of tools, to control toxic proteins inside the cell, which will pave the way for development of new and safe RIP-based anti-viral and anti-cancer targeted technologies.

ELIGIBILITY CRITERIA:

• Hold a master's degree in biology, molecular biology, biochemistry or related fields (not longer than 4 years).

• Have proven knowledge/experience in at least one of the following fields: molecular biology, biochemistry and/or structural biology.

• Have experience/knowledge in protein expression and purification; knowledge of structural studies (cryo-EM, X-ray crystallography) will be considered as valuable advantage.

• Have knowledge/training in various techniques including: gene cloning, DNA/RNA agarose electrophoresis, SDS-PAGE, PCR and other DNA/RNA/protein handling techniques.

• Publications in refereed journals, any internships abroad, or teaching abilities will be considered as additional advantages.

• Great work capacity and enthusiasm for research.

• Strong motivation to undertake experimental work and new scientific tasks.

• English with a good level of communication.

• Any nationality or gender.

• The candidate will be obligated to join or should have the status of a doctoral student at UMCS Doctoral School, at the beginning of participation in the project and during the implementation of tasks in the project.

WORK TASKS:

The project is focused on molecular and cellular activity of the ribosome inactivating proteins (RIPs). The PhD student will be involved in research encompassing work in the field of biochemistry, biophysics and structural biology (especially, cryo-EM) to gain an insight into molecular nature of interactions between the RIPs and eukaryotic ribosomes. The special tasks for the PhD student include: designing of the genetic systems for heterologous expression of various RIPs, recombinant protein purification, and study protein-protein interactions in the designed biological systems using the hi-end technology; the work will include also preparation of protein complexes for subsequent structural analyses using cryo-EM approach.

The PhD student will have the opportunity to work in the international scientific community, attend international conferences and training; and will be able to interact with leading experts through the network of academic international collaborations.

 

Specific Requirements

•  Curriculum vitae containing information on the candidate's scientific achievements (including possible participation in research projects, authorship and co-authorship of publications, awards, scholarships, scientific experience gained in the country and abroad).

•  A copy of a master's degree diploma or an official document confirming obtaining a master's degree, plus very short description of your Master thesis subject.

•  Copies of documents/certificates confirming professional qualifications.

• Candidate's declaration of knowledge of the English language or certificate.

• A short declaration of intention to join to the Doctoral School of Quantitative and Natural Sciences, UMCS in Lublin or a certificate confirming the status of a doctoral student at the Doctoral School.

• Opinion of the scientific supervisor/s.

WE OFFER

• Employment period: maximum 4 years (48 months).

• Scholarship in the amount of at least PLN 4,000/month gross (up to PLN 5000),

• Place of task implementation: Department of Molecular Biology, Institute of Biological Sciences, Maria Curie-Skłodowska University in Lublin

• Planned start: November 2023.

The scholarship is granted according to REGULATIONS FOR AWARDING NCN SCHOLARSHIPS FOR NCN-FUNDED RESEARCH PROJECTS.

The NCN research scholarship may be combined with other scholarships and salaries financed from NCN funds awarded under more than one research project, with the reservation that the total amount of scholarships and salaries collected may not exceed 5 000 PLN per month.

Additional comments

The best candidates will be invited for short job interview that will be organized on the online platform.

The competition may not be settled due to lack of candidates meeting the requirements.

The decision of the Recruitment Committee cannot be appealed.

As part of the competition procedure, we reserve the right to invite only selected candidates for an interview, as well as the right not to settle the competition or to cancel it.

In the case of the selected candidate refuses to accept the scholarship, it will be awarded to the next person on the qualifying list.

11. The data subject has the right to lodge a complaint with the President of the Office for Personal Data Protection, if he/she considers that the processing of your personal data violates the provisions of the GDPR.
