The effect of endocrine disruptors on the viability of human spermatozoa in vitro

Abstract
In the environment there is a whole range of compounds that have the ability to disrupt or completely alter endocrine processes in animal systems. These are natural substances but also a whole range of compounds of anthropogenic origin. These substances or their metabolites get into the food chain and can cause damage to a certain system. In addition to the known and proven effects of chemical compounds that are considered endocrine disruptors (bisphenols), in the presented work we will also monitor the potential negative effect of nanoplastics.
The work will be focused on the determination of damage to human germ cells due to the influence of different concentrations of bisphenols and nanoplastics in in vitro conditions and the possible elimination of the harmful effect. Determination of the mechanism of effect of the observed disruptors at the level of physiological parameters of sperm motility, membrane and acrosome integrity, mitochondrial activity and the level of oxidative status or. of the redox status of intracellular signaling pathways. The processing of the results, statistical analysis and gradual interpretation of the results will support the proposed hypotheses of the work, and the formulated conclusions will be presented at the domestic or foreign scientific conferences and professional symposia
Scientific goals: (1) identification of the activation mechanisms of cells against oxidative stress (biochemical, genomic, proteomic), (2) activation of protective and antioxidant genes during the response of cells to the resulting stress, (3) identification of structural damage of experimental cells.
Methodological objectives: Cultivation of sperm with bisphenols, nanoplastics in different concentrations. Determination of CASA motility parameters and MTT viability, IFA membrane integrity. The work will use the equipment of the Laboratory of Cellular and Subcellular Structures of the Institute of Applied Biology, Laboratories of Experimental Biology of RC AgroBioTech and partner foreign institutions.
Expected benefits: (1) determination of motility parameters of human spermatozoa after in vitro exposure to bisphenols and nanoplastics, (2) in vitro detection of sperm viability after exposure to different concentrations and time periods of bisphenols and nanoplastics, (3) influence of bisphenols and nanoplastics on oxidative and antioxidant status of human spermatozoa The result of the work will be clarification of the effect of selected endocrine disruptors and nanoplastics on the viability of human spermatozoa in vitro and the eventual possibility of eliminating their effect.
Vplyv endokrinných disruptorov na viabilitu humánnych spermií in vitro
Abstrakt
V životnom prostredí sa nachádza celá škála zlúčením, ktoré majú schopnosť narušiť resp. úplné meniť endokrinné procesy v živočíšnych systémoch. Jedná sa o prirodzené látky ale aj o celú škálu zlúčenín antropogénneho pôvodu. Tieto látky resp. ich metabolity sa dostávajú do potravového reťazca a môžu vyvolávať poškodenosť určitého systému. Popri známych a overených účinkoch chemických zlúčenín, ktoré sa považujú za endokrinné disruptory (bisfenoly), v predkladanej práci budeme sledovať aj potencionálny negatívny účinok nanoplastov.
Práca bude zameraná na determináciu poškodenosti humánnych pohlavných buniek vplyvom rôznych koncentrácií bisfenolov a nanoplastov v in vitro podmienkach a prípadnú elimináciu škodlivého účinku. Určenie mechanizmu účinku sledovaných disruptorov na úrovni fyziologických parametrov pohyblivosti spermií, membránovej a akrozómovej integrity, mitorchondriálnej aktivity a urovne oxidatívneho statusu resp. redoxného statusu intracelulárnych signálnych dráh. Spracovanie výsledkov, štatistická analýza a postupná interpretácia výsledkov podporia navrhované hypotézy práce a formulované závery budú prezentované na domácich resp. zahraničných vedeckých konferenciách a odborných sympóziách
Vedecké ciele: (1) identifikácia aktivačných mechanizmov buniek voči oxidatívnemu stresu (biochemická, genómická, proteomická), (2) aktivácia protektívnych a antioxidačných génov pri odpovedi buniek na vzniknutý stres, (3) identifikácia štrukturálneho poškodenia experimentálnych buniek.
Metodické zámery: Kultivácia spermií s bisfenolmi, nanoplastami v rôznych koncentráciách. Stanovenie parametrov motility CASA a viability MTT, membránovej integrity IFA. V práci sa využije prístrojové vybavenie Laboratória celulárnych a subcelulárnych štruktúr Ústavu aplikovanej biológie, Laboratórií Experimentálnej biológie VC AgroBioTech a partnerských zahraničných inštitúcií.
Očakávané prínosy: (1) stanovenie parametrov motility humánnych spermií po in vitro pôsobení bisfenolov a nanoplastov, (2) in vitro detegovanie viability spermií po účinku rôznych koncentrácii a časových periódach bisfenolov a nanoplastov, (3) vplyv bisfenolov a nanoplastov na oxidačný a antioxidačný status humánnych spermií Výsledkom práce bude ozrejmenie vplyvu vybraných endokrinných disruptorov a nanoplastov na viabilitu humánnych spermií v in vitro a prípadná možnosť eliminácie ich účinku.
