Offer Description
The PhD student will be part of a new collaboration between (SOPhy- INC-UMR 7177-http://sophy.chimie.unistra.fr) and IBMP, (INSB-IBMP CNRS UPR2357). He/she will share his/her time between the two teams. The synthetic part of the project will focus on the development of innovative methods to synthesize novel molecular tools dedicated to the visualization of phytosterols in plant cells. With regard to biochemistry and biology, he/she will contribute to setting up bioassays using model plant systems. These assays aim to map functional phytosterols in cells and organs. We are looking for a highly motivated, team-oriented candidate with good communication skills. The candidate must hold a Master's degree in organic chemistry. Solid experience in organic synthesis is required. Basic knowledge of biochemistry would be an asset but is not mandatory. Potential candidates should provide a detailed CV, M1 and M2 transcripts, contacts of their Master's internship supervisor(s), a cover letter and a brief summary of their Master's work (2-3 pages).
The project aims to develop an unprecedented set of molecular tools dedicated to phytosterol chemical biology facilitating the intracellular tracking of sterols to gain insights into their functions. Unlike mammalian and fungal cells, plant cells produce multiple sterols that exert diverse functions yet poorly characterized. The conjugation of new synthetic sterols to fluorophores via click chemistry will be used to visualize the incorporation of exogenously fed synthetic sterols. A key task, will be the regioselective introduction of an azide moiety into the sterol framework. A regio- and stereoselective azidation of sterols equipped with an N-allenamide is planned. Subsequently, a more challenging approach involving direct radical C-H azidation of C(sp3) bonds of sterols will be investigated, ensuring the production of structurally identical species to their native counterparts. Confocal microscopy will help tracking the intracellular localization of synthetic sterol in plants and provide valuable insights into their reaction mechanisms.
