Offer Description
The Nanobio team is developing new concepts to push back the limits of single molecule localization microscopy, in particular for live cell imaging. This position will involve developing a new concept recently patented for the localisation of individual fluorescent molecules. By encoding the position within a specific illumination, alternative original detection can be implemeted in particular to speed up acquisition, but also overcome the obstacles in terms of in-depth observation. In particular, the aim will be to design new optical implementations for setting up a chirped structured excitation (in 2D then 3D) offering uniqcity between the position and frequency of the detected signal, making it possible to use a single detector alone or in a matrix to analyse a wide field of observation. The aims of this project are to make it possible to observe/trace single molecules at previously unattainable acquisition rates, and also to open up new areas of observation in spectroscopy, by combining position information on a nanometric scale with detection of the spectrum or lifetime of fluorescence. These new developments are based on an initial patented proof of concept, and will be implemented as part of the TimeNanoLive ERC.
The post may be extended for a further 12 months.
The person will contribute to various aspects :
- setting up the optical implementations of this new concept
- testing and validation of the new experimental set-up
- pilot/simulation programming
- characterisation of various samples
- writing articles
- taking part in conferences
This work will be carried out within ISMO (Unité Mixte CNRS / Université Paris-Saclay) as part of a European proejct ERC TimeNanolive ). The laboratory has an on-site culture room and various microscopes for single molecule localization microscopy.
