Offer Description
The person recruited will take part in the "SULFOGLIA" project funded by the French National Research Agency (ANR). The project aims to understand the response and elimination of pathogenic substances by microglia, the brain's immune cells, in Alzheimer's disease. We will analyse the mechanism by which sulphated glycans present in microglia regulate their function, and the possibility that remodelling of sulphated glycans could improve clearance of pathogenic substances by microglia in the Alzheimer brain.
- Culture of primary microglia and microglia cell lines, and assessment of phagocytosis
- Maintenance of genetically modified mice, immunohistochemistry on cryo-cut tissues
- Production, molecular phenotyping and maintenance of lines modified by stable or transient cell transfection:
- Carrying out immunolabelling, immunoprecipitation, Western blotting and preparation of samples for proteomic and glycomic analysis, and transcriptome analysis by RNA-seq;
- Photonic and confocal microscopy analyses of fluorescent molecules;
- Drafting a manuscript, formatting and presenting results, keeping a laboratory notebook.
The staff recruited will work at the UGSF, UMR 8576, in Building C9 of the Faculty of Science and Technology, University of Lille. The "SULFOGLIA" project is funded by the French ANR.
This post will reinforce the research themes relating to the consequences of deregulations in the biosynthesis of heparan sulphates, which is being developed in the "Heparan sulphate diversity and inflammatory responses" team. This theme is part of a priority area for the UGSF on the involvement of pathological deregulations of glycosylation mechanisms in inflammatory and neurodegenerative pathologies.

Additional Information
- In-depth theoretical knowledge of glycobiology and cellular and molecular biology essential ;
- Practical skills in : (1) design and implementation of molecular biology and biochemistry techniques (RT-PCR, site-directed mutagenesis, cloning, sequencing, expression vectors, SDS-PAGE, immunoblot, etc.); (2) cell culture (maintenance of cell lines, selection of cells modified by stable and transient transfection, siRNA); (3) handling and use of genetically modified mice, organ harvesting, isolation and culture of primary cells (4) optical methods (microscopy, immunofluorescence), image acquisition and analysis. (5) bioinformatics: colocation analysis, heat map analysis, clustering, multidimensional scaling.
- A working knowledge of English is essential for understanding technical documents and scientific articles.
