Scientific context

In eukaryotic cells, gene expression is closely controlled by the dynamic modification of histones through the addition of covalent chemical groups on certain amino acids. The most well-known modifications are methylation and acetylation of histone lysine. Their diversity has greatly increased over the last decade with the discovery of a wide range of acylations, structures that are quite close to acetylation but which endow modified lysines with different physico-chemical properties. These acylations can be added to lysines from the corresponding acyl-CoA metabolites. A key question that has emerged from the discovery of these new epigenetic marks is whether they fulfill redundant functions with acetylation or have specific roles, particularly in the dynamics of chromatin structure and the control of gene expression. Answering this fundamental question would likely open new routes of drug development in the context of many diseases, to complement the existing panel of drugs that target the enzymes catalyzing the addition/removal of histone lysine acetylation or target the protein readers of these marks.

We are seeking to support a talented researcher in view of applying for an MSCA individual fellowship to contribute to unveiling the roles of newly described histone lysine modifications on gene expression regulation, by combining biochemistry experiments, proteomics, genomics and metabolomics. The successful post-doctorate would join Delphine Pflieger’s group who recently published a study on the crotonylation of histone H3 in the context of mouse spermatogenesis, using the above combination of omics technologies (Marion Crespo et al., Nucl. Acids Res. 2020).

Hosting description

This post-doctoral project would be carried out in the team Exploring the Dynamics of Proteomes (EDyP, www.edyp.fr). EDyP has developed mass spectrometry-based and computational strategies to finely and comprehensively characterize proteomes, in order to provide new insights in basic biology and translational research. EDyP is part of the Large-Scale Biology laboratory (BGE, http://www.bge-lab.fr/en) that brings together three research teams which implement large-scale strategies (proteomics, RNA interference, chemogenomics) to decipher the mechanisms controlling major cellular functions. Our laboratory is located within CEA Grenoble. CEA, the French Atomic and Alternative Energy Commission is a public government-funded research organization established in 1945. Through its nine established centers spread throughout France, CEA is a key player in research, development and innovation in four main areas: defense and security, low carbon energies (nuclear and renewable energies), technological research for industry, fundamental research in the physical sciences and life sciences (DRF). The key research axes of the DRF in the field of health are: improving knowledge in pathogenesis and translating and validating scientific advances into innovation for the patient. Health research is supported by a large technological expertise (“omics” technologies, structural biology, imaging, pharmacology and preclinical expertise) developed through several facilities and platforms integrated in National and European infrastructures.

WHO CAN APPLY

- 2-year research project in Grenoble, France. Possible stay in the laboratory of a European collaborator.

MSCA IF ELIGIBILITY CRITERIA:

- The researcher must be an experienced researcher (PhD or at least 4 years of full-time research after master's degree)

- The researcher may be of any nationality and can come from anywhere in the world

- The researcher cannot have resided or carried out his/her main activity (work, studies, etc.) in France for more than 12 months within the last three years

 

OFFER REQUIREMENTS:

- PhD in epigenetics or biochemistry; the candidate should have a strong interest in histone modifications.

- Languages: fluent English; optionally French
