OFFER DESCRIPTION

The person recruited will be assigned to the LCPME (UMR 7564 UL-CNRS) which includes 79 people distributed in three teams. He/she will join the Environmental Microbiology team on its theme "dissemination of antibiotic resistance in the environment" under the direction of Dr. Xavier Bellanger. The project to which the person will be assigned will take place in the context of a collaboration with an INSERM unit developing antimicrobial biomaterials and a start-up whose activity is focused on the development of phage therapy approaches.

The usual treatments of bacterial infections with antibiotics are facing more and more frequent failures due to the dissemination of antibiotic resistance. In this context, phage therapy, or the therapeutic use of bacterial viruses, called bacteriophages or phages, to fight infections caused by antibiotic-resistant bacteria is a promising therapeutic approach. However, considering that it could be implemented on a medium or large scale requires that this approach benefits from developments and improvements in terms of ease and modality of administration, safety and efficacy.

With the ANR PHAGERIALS project, we propose to focus on Staphylococcus aureus and Staphylococcus pseudintermedius, two staphylococcal species of nosocomial and zoonotic interest. PHAGERIALS aims to develop biocompatible hydrogel-based coatings / biomaterials that can be loaded with anti-staphylococcal phages. The PhD student (M/F) who will be recruited in the Environmental Microbiology team of the LCPME will be involved in this work by i) participating in the isolation and the genetic and physicochemical characterization of new phages, ii) rationalizing the selection methods of phages relevant for being used in phage therapy in order to limit the induced propagation of antibiotic resistance and pathogenicity genes, while ensuring the use of viruses with a broad spectrum of activity on target clonal complexes and, iii) actively participating in the development of hydrogels that can be easily applied to a surface or a wound, loaded with a high amount of virus while preserving their viability as long as possible, and that can release these viruses at the appropriate time, in the present case, during skin infections.
