OFFER DESCRIPTION

A first objective of the project will be to extract and purify the membranes of the purified capsids in order to identify the membrane proteins (viral and cellular) that compose it. In vitro activity tests will be developed to identify the fractions that may contain the assembly machinery. Finally, extraction protocols will have to be developed in order to study these complexes by electron microscopy. Complementary proteomics analyses will provide targets that can be expressed and purified and standard methodologies (nanodiscs, crystallization, Cryo-EM) are used to resolve the structures of individual proteins and pre-assembled complexes in vitro.

The proposed project aims to elucidate how the Mimiviridae genome is assembled in the icosahedral capsids. Since the disorganized DNA must be transferred through at least one lipid membrane and finds itself compacted in a helical structure of 30nm section, specific machinery must be dedicated to this process. The final goal of this project will be to compare this assembly machinery with the DNA translocation machinery used in the cellular and viral world.

The recruited person will be attached to the IGS laboratory - UMR7256.
Some long experiments may necessitate to adapt the working time to these constraints (night, weekend) if necessary.
The IGS laboratory is a pioneer and world leader in the study of giant viruses. We participated in the discovery of Mimivirus, the first giant virus, and discovered three other known families. The strength of the IGS laboratory lies in the coexistence of a team of bioinformaticians and a team of experimentalists, allowing to go from the isolation of new microorganisms to their complete characterization by combining approaches including genomics, transcriptomics and proteomics. The laboratory also has a strong expertise in structural biology allowing to determine the 3D structures of viral proteins of interest by crystallography and by combining crystallography and electron microscopy. The laboratory is equipped for cell biology (1 L2, 6 PSM, 3 fluorescence microscopes including 1 Zeiss Axio); biochemistry (PCR, 1 PSM Microbiology, 3 incubators, 2 Aktä explorer 10S, 1 Aktä 3D, 1 Aktä Prime, Nanodrop, Qubit); structural biology (Mosquito crystallization robot, a diffractometer (Oxford diffraction Xcalibur PXUltra, a ThermoFischer speedvac SPD131). The IGS has access to 12 technical platforms of the Institute of Microbiology of the Mediterranean (IMM, AMU-CNRS FR3479, Staff: 300) including a fluorescence and electron microscopy imaging platform (TEM, Cryo-TEM), NMR (600 MHz, cryoprobe), Proteomics, Protein Expression and Interaction studies (1 Aktä prime, 1 Aktä Pure, 1 BLItz, ITC, Biacore). The iGS also has access to the imaging platform of the Luminy campus (TEM and STEM) IBDM (Institut de Biologie du Développement de Marseille). The IGS belongs to the BAG SUD and has access to synchrotrons (ESRF, Soleil), electron microscopy platforms (Titan Krios cryo-electron microscope, K2 Summit direct electron detector, Quantum LS imaging filter and a Volta phase plate VPP). These microscopes on the line (CM01) (PSB) and at the IBS and EMBL. The IGS collaborates with the Kay Grünewald laboratory, CSSB (Hamburg) equipped with 5 cryomicroscopes including 2 Titan-Krios (300 keV top end, autoloader, DED, energy filter, VPP). The IGS has the necessary computing power (71Node cluster, 156 CPU, 828 Cores, 4700 GB, 150 TB).

Strong expertise in Structural Biology and solid experience in membrane proteins: Biochemistry, Cryo-Electron Microscopy, Crystallography. English (B1 to B2 level)
