OFFER DESCRIPTION

This interdisciplinary project will benefit from the stimulating collaborative environment of the Bio group at CRPP, between the Soft Micro Systems team led by J-C. Baret, expert in microfluidics and the Biopiles and Biosensors team led by N. Mano, expert in biosensors.

The activities within this project will involve:
- Microfluidic chip designs
- Microfabrication of molds and chips by 'soft-lithography' and 'replica molding' processes
- Testing and development of microfluidic chips
- Instrumentation, experiment set-up: encapsulation, microfluidic droplet manipulation, fluorescence and absorbance measurements
- Data collection and analysis, statistics
- Reports, presentation and scientific communication

Continuous blood glucose monitoring increasingly contributes to better health and quality of life for diabetic patients. However, the majority of implantable enzymatic biosensors are still facing with a loss of measurement precision over time due, among other things, to the inflammatory reaction.
The WEB project aims to develop a completely new strategy with a dedicated implantable electrode-antenna integrating an enzymatic glucose biosensor, an energy source and wireless data transmission. Based on an already active collaboration between four Bordeaux teams of electronics engineers, electrochemists and biologists, the results of this 4-year project will be validated by implantations on rodents.
The main objective of our work will be the optimization (activity/thermostability) of the enzymes used to create such a biosensor. To achieve this goal, we will use a directed evolution approach in microfluidics. Based on systems already developed in the laboratory, we will optimize microfluidic sorting systems allowing the screening of large libraries of enzyme variants. These variants will be expressed in bacteria and then in yeast allowing both single cell screening and the production of purified proteins.

The Paul Pascal Research Center (http://www.crpp-bordeaux.cnrs.fr/) is widely recognized for the development and characterization of complex systems and covers scientific fields ranging from the physics and physical chemistry of soft matter to the frontier with biology and the development of functional materials.

Eligibility criteria

The candidate must hold a Ph.D in physical-chemistry, physics or engineering for less than two years, skills and knowledge in the implementation of microfluidic droplet systems. Skills/knowledge in molecular biology and enzymology will also be appreciated. He/she will have a strong taste for technology transfer and for working in a multidisciplinary field. The candidate must be rigorous, have excellent organizational and teamwork skills, have the ability to lead, and be able to express himself / herself in English, written and spoken. He/she should have good communication skills allowing him/her to be integrated in the scientific activities and the life of the laboratory.
