OFFER DESCRIPTION

- Perform cloning and site-directed mutagenesis experiments
- Express recombinant proteins in mammalian cell lines
- Purify proteins using FPLC systems and perform their quality control
- Optimize the production of various protein complexes for cryo-electron microscopy (cryo-EM)
- Participate in cryo-EM data collection and analysis (in collaboration)
- Carry out protein-protein interaction studies by SPR or thermophoresis
- Perform biochemical assays to measure/compare protein amounts by Western blot, ELISA or trageted proteomics (PRM)
- Check published litterature in the field
- Make oral and written reports (project meetings, group meetings, conferences etc)
- Participate to common lab tasks (cell culture room cleaning, consumable stock monitoring etc)

The selected candidate will perform biochemistry and structural biology experiments in relation with the proteolytic complexes involved in collagen maturation and fibrillogenesis. He/she will work with different types of samples including purified proteins, cell cultures, plasma and tissue biopsies.

The work will mainly take place at UMR5305 (LBTI) in Lyon Gerland but short stays at the OPIC (Oxford Particle Imaging Center) facility of the STRUBI lab in Oxford are planned. LBTI is a pluridisciplinary laboratory with interest in tissue biology (homeostasis, stress response, aging etc) and therapeutic target identification. The host team works on extracellular metalloproteases and extracellular matrix in the context of tissue repair and fibrosis. The project will include biochemistry, structural biology but also analysis of human and murine samples. The selected candidate will have access to the cutting-edge facilities of the SFR Biosciences and the SFR Santé. The project is funded by the French Research Agency (ANR).

Eligibility criteria

- Excellent theoritical and practical knowledge of molecular biology and protein biochemistry
- Experience in running protein-protein interaction analyses (ideally SPR) and biochemical assays to measure protein concentrations (ELISA, targeted mass spectrometry)
- Knowledge in structural biology; previous experience in cryo-EM would be an advantage
- Knowledge in extracellular matrix bology
- Fluency in English (collaboration with a group in Oxford)
- Experience in writing scientific articles

The post-doctoral position can be a stepping stone to become a permanent CNRS researcher (competitive exam)
