The work will take place in the GEPHIR project (Glycan epitope profiling by hyphenation of IR laser spectroscopy and separative methods). We propose an analytical development for glycomics, a field which lags far behind genomics or proteomics. The method will aim at the analysis of glycan epitopes (acetylation of sialic acids and sulfatation of terminal sugars) which is out of reach of current analytical methods and hinder the full characterisation of the glycosylation profiles of glycoproteins. The original concept proposed here lies on the coupling of IR ion spectroscopy with several dimension of upstream separation ( LC, ion mobility, MS) to resolve the isomeric patterns present in complex heterogeneous samples. The project brings together a consortium of acknowledged teams in glycomics, ion spectroscopy, glycochemistry and immunology. A demonstration of the potential impact of the method will be made through a case study on the early diagnostic of rheumatoid arthritis in collaboration with the public hospital in Lyon.
The work will take place at Institut Lumière Matière and will consist of the analysis using the MS/IR coupling, and the implementation of ion mobility.
We're looking for a candidate with a Master degree (M2) in molecular sciences or analytical chemistry. Experimental skills, as well a s-a good motivation to work in an interdisciplinary team are required.
