Fightingmuscle atrophy by inhibitingtheUbiquitin Proteasome System (UPS)

A feature of several diseases (cancer, sepsis, heart failure, ...) is a catabolic state leadingto muscle wasting. This muscle wasting is mainly due to a huge increase in skeletal muscle proteolysis, which, if sustained, leads to muscle weakness, compromises treatments and is associated with increasedmortality.The project presented here focuses on the main proteolytic systeminvolved in these cases, the UPS, that specifically degrades proteins taggedby a polyubiquitin chain. Polyubiquitination and selective targeting are ensured by pairs of enzymes calledE2-E3 couples. In muscle,the degradation ofcontractile proteins involves an E3 ligasenamed MuRF1. The first partof the projectis to specify the E2 enzyme working with MuRF1 to target contractileproteins, thus leading to their degradation and therefore to skeletal muscle atrophy(subcellular co-location.Interactomic of E2-MuRF1 in presence of substrates. In vivoknock-out and overexpression studies).Secondly, inhibitory molecules will be screenedand characterized in vitro and in vivo. This topic is notably supported by a Marie Skłodowska-Curie Innovative Training Network (ITN)2019-2022 funding,benefiting from aEuropeannetwork.
