Indispensable amino acid restriction as a preventive nutritional strategy: role of theGCN2-ATF4 signalling pathwayregulationby plant proteins

The regulation of gene expression by amino acidavailability is a fundamental mechanism allowing the organismto adapt its metabolism toface thevariations in amino acid intake and in particularinindispensableamino acids (IAA)intake. The signallingpathway involving the kinase GCN2 and the transcription factor ATF4 plays a major role in the adaptation to nutritional restriction in IAA. Activation of the GCN2-ATF4 pathway has been shown to exert a preventive effect in various stressful situations. In parallel, other studies have shown that a methionine restriction, an IAAI, improves metabolic parameters in obesity models. However, a permanent restrictionin IAA is not possible over a long periodin human. In this project, we propose to develop a nutritional strategy in mice to induce an intermittent activation of the GCN2-ATF4 pathwayover a long period. This nutritional strategy is based on the use of plantproteinsthathave anunbalanced amino acid compositionincluding lysine or methionine. Then, the objective will then be to evaluatewhether this nutritional strategy is able to exerta preventive role in a model of induced obesity. Through different bioluminescent and KO transgenic models, the role of the GCN2-ATF4 pathwaywill be investigated.
