We are looking for two candidates to start two Joint PhD research projects between the Université de Sherbrooke and the Université Paris-Saclay. These two projects aim to study the interactions between bacteriophages (i.e. viruses or phages) and the pathogenic bacterium Clostridioides difficile, which is the main cause of post-antibiotic diarrhoea and is one of the most problematic bacteria in hospitals.
The first project will focus on the study of an antiphage system of the "abortive infection" (Abi) type controlled by a riboswitch that has been identified in the genome of a prophage. This project will be carried out mainly at the Université Paris-Saclay under the supervision of Prof. Soutourina, but will include a significant portion (at least 1 year) of work at the Université de Sherbrooke in the laboratory of Prof. Fortier.
The second project will focus on the study of the binding specificity between phages and their bacterial receptor, more specifically the Surface-layer protein A (SlpA). This project will take place mainly at the Université de Sherbrooke in Quebec under the direction of Prof. Fortier, but will include a significant portion (at least 1 year) of work at the Université Paris-Saclay in the laboratory of Prof. Soutourina.
For both projects, the research activities will include:
- Anaerobic culture of C. difficile and infections with bacteriophages in a biocontainment level 2 working environment
- Molecular cloning (construction of plasmids and mutants of C. difficile using different genetic and molecular tools)
- Protein expression in C. difficile
- Phenotypic characterization of C. difficile (growth, toxin production, sporulation)
- Gene and protein expression analysis (Northern blot, RNA-seq, RT-qPCR, Western blot, reporter gene fusion)
Candidates should have a Master 2, MSc or equivalent degree. Candidates should ideally have relevant experience in microbiology and/or molecular biology, have good communication skills, and be prepared to study abroad for several months.
