The Fellowship implies carrying out the activities briefly mentioned below at Dipartimento di Scienze Cliniche e di Comunità dell’Università degli Studi di Milano and under the supervision of Prof. Sergio Abrignani within the research programme “Isolation and phenotypic characterization of lymphocytes infiltrating the tumor”, within the research project n. CP2_12/2018 financed by Fondazione Regionale per la Ricerca Biomedica entitled “A regional oncology network addressing the emerging problem of colorectal cancer in young individuals using an integrative omics approach to decipher mechanisms of cancer immunoediting as candidate targets of new therapies”, of the activities mentioned below:
1) Isolation and phenotypic characterization of lymphocytes infiltrating the tumor.
2) Functional characterization of lymphocyte subsets and transcripts identified by single-cell transcriptomics.
REQUIRED EDUCATION LEVELMedical sciences: Master Degree or equivalent
Expertise in research area: previous research experience in the field of immunology, cell biology and molecular biology. Purification and culturing of lymphocytes isolated from tumor tissue and from peripheral blood. Multiparametric cytofluorimetry for phenotypic and functional analysis of tumor infiltrate (cell cycle, intracellular staining on fixed cells, cell viability and apoptosis, proliferation). Molecular biology techniques (Single-cell RNA-Seq, PCR, RT-qPCR, design and implementation of cloning). Phenotypic and functional study of genes and proteins. ELISA and high density array tests for cytokine dosing in biological fluids. Basic microscopy techniques (immuno-fluorescence, IHC). Familiarity with flow cytometry analysis software (FlowJo). Development of cellular assays to study the interaction between immune system and cancer cells.
