The position is available as of now. The position is funded according to § 2 WissZeitVG and § 65 HHG with the opportunity of own scientific training. The salary of the position is determined by payment group E13 according to the collective labor agreement of the State of Hessen (TV-H).

Scientific employees are – unless the allowed maximum duration of a temporary employment is exceeded – generally employed for an initial duration of 3 years with a possibility of extension for an additional 2 years, if the above-mentioned requirements are fulfilled.

Malaria parasites massively alter the biochemical and biophysical properties of their chosen host cell, the mature human erythrocyte. This host cell modification is driven by parasite-encoded proteins which are sorted to various localisations within the infected host cell. As this process is unique for malaria parasites, it is an attractive target for development of the next generation of anti-malarials, however we still lack a thorough understanding of these transport processes and the molecular mechanisms underlying protein trafficking in this system. This project aims to identify both signals and mechanisms required for the targeting of the parasite protein PfExp1 to the membrane of the parasitophorous vacuole. Techniques used will include reverse genetics (including CRISPR/Cas technology) both fluorescent and electron microscopy, and protein biochemistry.

Description and tasks:

• Own scientific further education and university didactic qualification; scientific service in research and teaching according to §65 HHG, especially collaboration in current research projects of the Professorship.

• Assumption of teaching duties in accordance with the Teaching Obligation Ordinance of the State of Hesse

• Conduct malaria cell culture (S2 laboratory)

• Fluorescence microscopy and immunofluorescence

• Production of mutants of the malaria pathogen Plasmodium falciparum using CrispR Cas9. Cloning of the constructs by PCR or gene synthesis. Determination of IC50 and Ki values of various inhibitors on these mutants

• Guidance of master students in the execution of their experimental master thesis

Requirements:

• Completed Masters or equivalent University degree in Biochemistry, Biology or related subject

• Willingness to work in a highly interactive and interdisciplinary team

• Interest in tropical infectious diseases

• Fluent in written and spoken English

• Strong background in cell culture, microscopy and molecular biology
