About Organovir

Organovir is an Innovative Training Network (ITN) that focuses on the use of organoid technology for viral pathogenesis studies and anti-viral testing. For this ITN we will provide 15 Early Stage Researchers (ESRs) with the opportunity to be trained within a consortium of public and private experts that will enable them to lead innovation in the field of organoids for virus research. The starting date of the research projects is 1st of September 2019.

 

As an ESR with Organovir, you will be able to familiarize yourself not only with the scientific and technical aspects of innovative 3D culture systems for virus research, but also with the financial, commercial, ethical, communicative, policy and dissemination aspects enabling optimal valorisation of organoids. At the end of this project, you will hold a unique position within the European labour market.

 

Within Organovir research will be done in the areas of virology, organoid technology, science ethics, communications and business. Organovir is therefore looking for talented and motivated ESRs with a Master degree in various disciplines depending on the research project you want to apply for (see vacancies for requirements).

Project title: Complex HAE model for studies on coronavirus pathogenesis (ESR 1)

Host institution: Jagiellonian University, Poland. The research will be carried out in a young and dynamic team lead by Krzysztof Pyrc. The Jagiellonian University is located in the historical, yet vibrant city of Krakow.

 

About the host institution

The team of prof. Krzysztof Pyrc works at two faculties: Faculty of Biochemistry, Biophysics and Biotechnology (FBBB) and Malopolska Center of Biotechnology (MCB). The first unit is teaching/training-oriented, the second unit (with high-containment laboratories) functions as a R&D unit, with state of the art facilities. FBBB JU is the best biology/biotechnology faculty in Poland, according to the 2017 governmental scientific unit assessment. The MCB is the first University – based research institute and at the same time the excellence center. It gathers excellent internationally renowned scientists and laboratories (including Max Planck laboratory and CNRS), and periodic evaluation of researcher coordinated by the independent, international advisory board ensures high scientific quality.

 

The focus of the research group is on the understanding of the infection process of human viruses and the interplay between the virus and the host cell, and translation of these results to the applicatory field. They have recently developed several virus inhibitors, some are in the stage of (pre-) commercialization (HTCC compound for animal coronaviruses, NSPAH polymer as broad antiviral compound, www.virogenetics.info). The teams’ expertise with human 3D cell culture models encompasses mainly human airway epithelium (HAE) cultures formed by primary human epithelial cells on an air/liquid interphase. They have successfully cultured type I and type II pneumocytes. Clinical material preparation (human lungs), cell isolation and differentiation are carried out in house. Further, the methods allowing for modification of 3D cultures (lentiviral gene engineering, gene silencing, co-cultures) and analysis (confocal microscopy, 3D virtualization, SEM, superresolution microscopy) allow to study viral infection in the context of natural tissue.

 

Project description

This project focuses on novel 3D complex model system to study infection with respiratory viruses. During this project, you will be focussing on the following research objectives:

  • To create a model for human airway epithelium allowing to study the interplay between the virus and our immune system.
  • To test what happens during coronaviral infection in such a model.
  • To understand what happen on a single cell level. Big data.

 

With this project, we aim to achieve the following results:

  • Develop complex HAE tissue, combining different cell types to reflect the interaction between the pathogen and the immune system.
  • On tissue level: map coronaviral infection, cell-to-cell spread, potential of immune cells as carriers, pathogenesis.
  • On single cell level: analyse the microenvironment of the cell, innate immunity, virus-cell interaction, virus specificity.

 

This project also includes two secondments, which are as follows:

  • A two-month secondment with Danmarks Tekniske Universitet (location: Lyngby, Denmark) focused on novel biomaterials / solutions for advanced 3D cultures.
  • A three-month secondment with the Universiteit van Amsterdam (location: Amsterdam, the Netherlands) focused on bringing gut organoids to JU and bringing complex HAE cultures to UvA.

 

You will be guided during your research project by Krzysztof Pyrc (Virogenetics Laboratory of Virology).

 

Requirements

Organovir is looking for research candidates who are highly motivated and enthusiastic with a flexible, pro-active team spirit and a recent Master degree in the field of biochemistry, cell biology, molecular biology, virology, tissue engineering, life sciences and related. Interest and experience in virus culture and organoid technology is an advantage. The candidate is required to be proficient in the English language and should be in the first four years (full-time equivalent) of their research careers. In addition, the applicant should not, at the time that he/she is selected by the host institution, have resided or carried out their work and/or studies in the country of the host institution for more than 12 months in the 3 years immediately prior to their recruitment. What we offer

We offer a 4-year research position for 40 hours per week within a motivated and innovative consortium. The starting date of the research project is the 1st of September 2019.

Eligibility criteria

You are eligible to apply for an ESR position if you meet the following conditions:

 

Early-stage researchers (ESR) are those who are, at the time of recruitment by the host institution, in the first four years (full-time equivalent) of their research careers. This is measured from the date when they obtained the degree which formally entitles them to embark on a doctorate, either in the country in which the degree was obtained or in the country in which the research training is provided, irrespective of whether or not a doctorate was envisaged.

 

Conditions of international mobility of researchers: Researchers are required to undertake trans-national mobility (i.e. move from one country to another) when taking up the appointment. At the time of selection by the host institution, researchers must not have resided or carried out their main activity (work, studies, etc.) in the country of their host organisation for more than 12 months in the 3 years immediately prior to their recruitment. Short stays, such as holidays, are not taken into account.
